With Arduino IDE which you must have for the B4R to work anyway.

.ino files are Arduino IDE files...
.b4r files are B4R IDE files...
When compiling in release mode, the B4R IDE converts (compiles) the .b4r file in to .ino and uses the Arduino IDE command line to upload the "sketch" to the board.
